1999 GTS Will Not Run On Its Own

I have a 1999 GTS that will not run on its own.
If I inject premix either into the cylinders or into the carburetor it will fire up. Then, it stalls out for lack of fuel.
I have replaced the fuel lines and have rebuilt the carburetor.
The carburetor is getting fuel for if I disconnect the fuel line, it'll pee.
I understand the initial setting for the high speed jet is 0 turns out. Should I crack this adjustment 1/8 turn out?
How do I know if the fuel pump side is working properly? Any bench testing I can do?
how do I know the puffer into the fuel pump is correct?

Where did you buy the carb rebuild kit from? You can test the fuel pump diaphragm with 4 pounds of air and see if it holds, don’t use a compressor to add the 4 pounds. Have you taken a compression test lately?
 
Source of kits not remembered. Packaging was Mikuni.
Compression taken simultaneously with separate gauges: #1, 130 psi taken with Sears / Craftsman gauge; #2, 135 psi taken with Lisle gauge.
I shall try to test for the 4 pounds of air.

You don't test compression with two different gauges.
One at a time with the same gauge, hold throttle wide open and crank until gauge stops climbing.
 
High speed wont affect starting, it is for the upper power band. the low speed can affect it slightly , but normally if you set them at factory you should be golden.
What i would do is take a hose and put it in premix gas and connect it to the carb "in side, see if it runs, if it does you have issues down the fuel line, if that is the case just keep adding the components until you get an issue. If this doesn't help it then you probably have a problem with the accelerator pump, I think your model has that, not 100% sure though.
